I have a question in my lab manual asking why when resuspending eukaryotic (mammalian to be precise) cells, a vortex machine cannot be used and it must be done gently with pipettes. Whereas with bacterial cells a vortex can be used.

I am assuming it is something to do with the cells bursting, or perhaps the nuclei bursting but cannot find any evidence online or in books to back this up. Does anyone know the answer??

You are correct in guessing about cells bursting- there is a large difference in cells size between eukaryotic and bacterial cells. The large cells are more fragile to forces and harsh treatment such as a vortex. Depending on your lab manual, it may also be due to the solution the cells are in.
